IN VITRO PROPAGATION OF GETHYLLIS MULTIFOLIA L. BOLUS (KUKUMAKRANKA) - A THREATENED SOUTH AFRICAN GEOPHYTE WITH MEDICINAL POTENTIAL

Gethyllis multifolia is a deciduous, bulbous geophyte that is indigenous to South Africa, is threatened and is listed in the ‘Vulnerable’ category of the ‘Red Data List of South African Plants’ and the ‘IUCN-World Conservation Union List of Plants’. This genus (including G. multifolia) has reported medicinal properties and the berry of this species is edible and has an extraordinary fruity flavour. No in vitro culture research has previously been done on this species and limited research, in general, has been conducted on this threatened genus. The objective of this research was to establish successful in vitro culture methods for the rapid multiplication of this species. G. multifolia explants were successfully sterilized in a 2% NaOCl and 0.1% Tween solution for 20 min with a contamination rate of less than 20% in the MS medium with the 8.1 µM NAA:1.8 µM BA growth regulator combination. No new shoots (only explant enlargement and callusing) were formed during the initiation stage, and a low percentage shoot proliferation was obtained during the multiplication stage and was similar (±20%) for the MS media supplemented with 0.5 µM NAA:14.7 µM BA, 5.4 µM NAA:0.9 µM BA and 8.1 µM NAA:0.9 µM BA growth regulator combinations. A rooting percentage of 83% was obtained on the MS medium supplemented with 2.7 µM NAA. Plant survival for rooted shoots was 88% in a medium with 1 sifted bark:1 perlite:1 sand (v/v), following transfer to plastic mini-tunnels in a greenhouse and watered by hand. The rooting and acclimatization stages proved to be successful but additional research is warranted on the growth regulator concentrations for shoot initiation and faster proliferation in this species. The existing conservation status and the expected pharmacological importance of this species emphasize the necessity of this research.
