DETERMINING ACCURATE HARVESTING TIMES OF COELOGYNE ASPERATA LINDL. SEED CAPSULES FOR PROPAGATION USING TISSUE CULTURE TECHNIQUE
Coelogyne asperata Lindl. is a beautiful orchid found growing on tree branches and rock faces near rivers in the primary forests of Borneo. As with many other beautiful native plants, it is no longer common in the wild as more of our forests disappear. Orchid is very difficult to grow naturally as the seeds do not have food reserves. Moreover, propagation by seeds is influenced by their maturity. The purpose of this study is to identify appropriate timing for harvesting seeds of Coelogyne asperata for propagation using tissue culture method, in order to reduce the failure during culture due to inappropriate seeds stage. The research was conducted at CV. Ayu Orchid tissue culture laboratory Bali, from November 2012- April 2013. Results shows that the optimal harvesting time for capsule of Coelogyne asperata is 4 months after pollination. The highest response for growth and higher number of explants is 4 month 14 days (208 seedlings), followed by 3 month 20 days (103 seedlings).
