CRYOPRESERVATION OF COELOGYNE DAYANUM SEEDS BY VITRIFICATION
The aim of the study was to investigate the effectiveness of vitrification technique for cryopreservation of Coelogyne dayanum seeds. Seeds from green capsules were precultured on solid media supplemented with sucrose at concentrations of 0, 0.3, 0.5, 0.8 M for 3 days and dehydrated in PVS2 solution for 0, 3, 5 and 8 h at 0°C before storage in liquid nitrogen for 1 week. Survival was determined through germination on Mitra basal medium containing 0.2% (v/w) sucrose, 2 g/L peptone and 1 g/L charcoal. Seedlings from cryopreserved seeds were cultured on half-strength MS or Mitra basal media supplemented with TDZ (0, 0.25, 0.50, 0.75, 1.00 mg/L) or kinetin (0, 0.50, 1.00, 1.50, 2.00 mg/L) or BAP (0, 0.50, 1.00, 1.50, 2.00 mg/L) for seedling development. Seeds precultured on the medium containing 0.3 M sucrose gave the highest survival of seeds post cryopreservation while dehydration in PVS2 for 3 h resulted in the highest germination rate (80.9%). Seedlings cultured on half-strength MS medium containing 0.5 mg/L TDZ promoted the maximum increment of seedling height (1.27 cm) and leaf length (0.52 cm). Seedlings cultured on Mitra medium turned yellow after 8 weeks of culture.
