The role of auxin and cytokinin on somatic embryogenesis from cell suspension cultures of the banana cultivar 'Cau Man'

This study examines the role of auxin and cytokinin on somatic embryogenesis from cell suspension culture of banana cultivar 'Cau Man' (AA genome, Sucrier subgroup). A highly proliferating meristem culture (CHP) of banana cultivar 'Cau Man' was initiated on CHP medium containing 0.17 mg L-1 indol acetic acid (IAA), 2.5 mg L-1 N6-benzyladenine (BA) and 1 mg L-1 zeatin. Multi-scalps formed by this culture were transferred to the liquid Ma2 medium with 35 g L-1 sucrose, 1.2 mg L-1 2,4-dichlorophenoxyacetic acid (2,4-D), 0.5 mg L-1 zeatin and 15 mg L-1 ascorbic acid to initiate cell suspensions. Roles of auxin and cytokinin on somatic embryogenesis from these cell suspensions were studied. Somatic embryos were obtained on Ma3 medium containing 0.1 mg L-1 kinetin, 0.2 mg L-1 6-dimethylallylamino purine (2-iP) and 0.05 mg L-1 zeatin, at initial cell density of 15 µL mL-1 SCV (settled cell volume). Cytokinin treatment increased the number of mitochondria observed under transmission electron microscope (TEM). IAA and zeatin levels in somatic embryos in different stages were determined by high performance liquid chromatography (HPLC). IAA level increased at the globular embryo stage. Use of TIBA (2,3,5-triiodobenzoic acid), an inhibitor of auxin polar transport, showed that auxin polar transport played an important role in the vertical axis formation of embryo organs as well as their development. The combination of auxin (1.5 mg L-1 IAA) and cytokinin (0.227 mg L-1 BA or 1 mg L-1 zeatin) at the volcano stage was necessary to complete the development and germination of somatic embryos. In LSQUOCau ManRSQUO somatic embryogenesis, IAA and zeatin, especially auxin polar transport, are major elements needed for embryo development.
