Advances in micropropagation of hazelnut (Corylus avellana L.) in Chile

The traditional methods of hazelnut micropropagation on a solid culture medium often present a low multiplication rate and a lower quality of the material with respect to the shoots obtained in the temporary immersion system. This study was carried out to determine the effect of the temporary immersion system on the multiplication rate in ‘Barcelona’ and ‘TGL 18’ (a selection of ‘Tonda Gentile delle Langhe’), and to obtain an efficient protocol for the rooting induction. In our research explants of 20-25 mm of ‘Barcelona’ and ‘TGL18’ have been used. The shoots have been maintained in a DKW inorganic salts together with pyridoxine 0.5 mg L-1, myo-inositol 100 mg L-1, glycine 2 mg L-1, thiamin 1 mg L-1, nicotinic acid 0.5 mg L-1, and sucrose 30 g L-1. The medium has been supplemented by BA 1 mg L-1 and IBA 0.01 mg L-1. The pH has been adjusted to 5.5 and 7 g L-1 of agar were added before autoclaving. The two cultivars showed that the temporary immersion system significantly enhanced the multiplication rate when compared to the solid medium that only reached a low proliferation rate. The most effective treatment in terms of rooting (100%) in ‘Barcelona’ and ‘TGL18’ resulted from the IBA and putrescine combination. For the temporary immersion system DKW liquid medium has been used without agar. The rooting was performed in DKW media supplemented with IBA (0, 0.5, 1, 1.5 and 2 mg L-1) and IBA + putrescine (1 mg L-1 and 1 mg L-1)
