New approaches to micropropagation of raspberry
The possibility of increasing the output of rooted microplants in cultivars of remontant raspberry, such as Herakl, Zhar-Ptitsa and Poklon Kazakovu was studied. Micropropagation was carried out on Murashige-Skoog medium with 6-benzylaminopurine (BAP) at a concentration of 1.0 mg L‑1. For rooting in the control, indolyl-3-butyric acid (IBA) was used at a concentration of 1.0 mg L‑1, in the experimental variant auxin was not added. Preliminary study of the plantlets rooting on the nutrient medium of Murashige-Skoog on the background of IBA 0.5-1.0 mg L‑1 revealed rhizogenesis in the range of 15-40% in all studied cultivars with low quality of the roots. In the passage preceeding the rooting, according to the traditional scheme, the concentration of BAP was reduced to 0.05 mg L‑1 and elongation was carried out in flasks with a volume of 250 mL to obtain plantlets of 2-3 cm in length. In the developed scheme, a separate stage of elongation was not envisaged. In the experimental variant, we combined the stages of elongation and rooting into one and from plantlets of 0.5-1.0 cm length for 1.5-2 months, we obtained rooted microplants, using also large-size cultivation vessels, namely, flasks with a volume of 250 mL. By the end of the second month of growth, the height of the stem part of the microplants of the experimental variant varied from 3.6 to 7.4 cm, and 70-95% of them formed roots. As a result, the rooting in the experimental variant was 2-3 times higher than the control, no separate subculture was required for elongation. It was possible to produce rooted, well-developed microplants within 1.5-2 months of initiation.
