Effect of genotype, stress conditions and media on androgenic performance of eggplant
This study was conducted with the purpose of investigation the effects of genotype, nutrient media, cold pretreatment and incubation treatment on haploid plant development in eggplant. Yamula, Karabaş F1, Malkara F1, Çantalı F1, and Tatlıcan F1 genotypes were used as donor plant.
Anthers were cultured in DDV and MS media with 0.01 mg L‑1 kinetin + 0.01 mg L‑1 2,4-D + 0.03 mg L‑1 vitamin B12. Flower buds were held at 4°C for 24 h.
After anthers were placed in the nutrient media, two different incubation treatments were done by holding them at 9 and 35°C for 8 days in the darkness.
Afterwards, anthers were held at 25±2°C for 16-h day/8-h night for 4 days and transferred to regeneration media.
While Malkara F1, Çantalı F1, and Tatlıcan F1 genotypes did not give any embryogenic response in all treatments, Yamula and Karabaş F1 genotypes developed embryos.
Moreover, the highest embryo development was obtained from Yamula. Embryo development rates varied by genotypes between 1.73 and 9.44%. While embryo development rate was 7.29% in DDV medium, it was 3.88% in MS medium.
Embryo development rate at 9 and 35°C incubation treatments were 0.27 and 10.90%, respectively.
As a result, haploid plant development was 0-92.30% in DDV medium, and 0-93.33% in MS medium.
