Study on asparagus tetraploid induction, ploidy identification and biological characteristics

The nutritional organs of polyploid plant are generally larger than that of diploid plant; the edible part of asparagus is immature stem, thus the polyploid breeding of asparagus has great significance for improving its production. In this paper, germinant seed and seedling of 'Jing Gang 701' diploid asparagus (a cultivar commonly planted in south China) as material, the comparison of soaking seed method dropping seed method on the induction effects of tetraploid was investigation. Firstly, the influence of the concentration of colchicine, the shaking speed of soaking, and the soaking time on induction of polyploids was investigated. The result showed that the best result can be obtained if 0.2% colchicine were used to soak germinant seed for 6 h, and the highest induction rate of tetraploid was 20%, but there was no significant difference in the influence of different centrifugation speeds on induction rate of tetraploid. Secondly, 0.3% colchicine was continuously dripped on the seedlings for 4 days, the highest tetraploid induction rate was 18%. It was the higher induction rate by soaking seed method compared to that by dropping seed method. Finally, this investigation also found that the tetraploid plant was stronger and also had stronger heat and drought resistance performance than the diploid plant.
