Optimization of protein extraction technique in walnut fruits
Protein composition was studied in order to determine the fractional structure of fruits kernels of 7 walnut cultivars (Alminsky, Arkad, Burlyuk, Dolinniy, Podarok Valentiny, Karlik 3, Karlik 5) of the Nikita Botanical Gardens selection grown in soil and climatic conditions of the foothill Crimea. Electrophoresis was carried out in a polyacrylamide gradient gel (6-12%). During the study of the analyzed plant material, we found significant differences for a group of proteins with molecular weights from 5 to 25 kDa. This is typical for fruit samples of three cultivars of walnut Podarok Valentiny, Dolinniy, and Alminsky, which exhibit significant differences in the analyzed indicators both in the profile of the molecular weights of proteins and in their quantitative content. The obtained results indicate the possibility of using the protein profile as an identifier for the cultivar of analyzed plant material. Further investigations are planned to study the amino acid sequence of proteins in the light fraction of proteins, and also to establish the level of their expression in different cultivars.
