Marker-free tomato plants expressed the supersweet protein thaumatin II gene

In our work previously we used the pMF1 vector containing the recombinase R and a CodA-nptII bifunctional selectable gene to produce tomato plants carrying the supersweet thaumatin II gene from a tropical plant under the control of fruit-specific tomato E8 or ELIP gene promoter and the tomato rubisco gene terminator. After Agrobacterium transformation delayed strategy was followed for the selection of marker-free transgenic tomato plants. One fully marker-free transgenic line was obtained. With the PCR analysis we confirmed that one completely marker-free transgenic tomato plant was obtained. The thaumatin II gene expression has been detected by RT-PCR in the primary transgenic plants as well as marker-free tomatoes.
