Biotechnological methods of reproduction and preservation of species and cultivars of the genus Syringa L.

The work represents the results of the research on multiplication and conservation of one of the most representative in Russia in vitro Syringa L. collection of Tsitsin Main Botanical Garden. Nowadays, the collection consists of 257 taxa (19 species and subspecies and 238 cultivars). It includes species samples from most series of the genus and also represents the most significant achievements of national and foreign lilac selection. During the research, the plant morphogenesis under impact of the nutrient medium components was studied. The optimal conditions at main cultivation stages for effective lilac reproduction and conservation were found. The half strength Murashige & Skoog medium was optimal for Syringa species reproduction. Quorin-Lepoivre medium stimulated shoot proliferation of lilac cultivars better than Murashige & Skoog medium. The combination of cytokinins and auxins as media additives at the multiplication stage showed better results than using only cytokinins. The modified Quorin-Lepoivre medium with combined addition of 1.0 mg L‑1 6-benzylaminopurine and 0.01 mg L‑1 indole-3-acetic acid as the growth regulators was optimal for obtaining high reproduction coefficient of most studied lilac cultivars. The storage culture conditions determined through our research allowed preserving lilacs for more than 2 years without transferring to fresh medium. Chlorocholine chloride at a concentration of 0.2-0.4 mg L‑1 as medium contribute was found effective for increasing the subculture period of long-term preservation.
