MICROPROPAGATION OF APPLE ROOTSTOCKS

Meristems with 2 leaf primordia were isolated from three apple rootstock buds (M 26, M 27 and MM 106) after the winter period. The meristems, cultivated on MS (Murashige and Skoog, 1962) and QL (Quoirin and Lepoivre, 1977) culture medium supplemented by BAP (0.9–1.5 mg l-1) formed leaf rosettes after 6 weeks. Shoot multiplication was achieved on both media with (mg 1-1): BAP 0.9, IBA 0.1 and GA3 0.1. In this phase both the effect of agar concentration and isolate position on the number of newly formed shoots was tested. The shoots were rooted on either half MS or half QL medium with sucrose concentration 1% and four different IBA concentrations (0.5, 1.0, 2.0 and 50 mg 1-1).
