APPLICATION OF DOUBLE-PHASE CULTURE MEDIA IN MICROPROPAGATION OF HYACINTHUS ORIENTALIS CV. CARNEGIE. II. EFFECT OF AUXINS ON GROWTH AND DEVELOPMENT

A. Bach, A. Cecot
In the search for an optimal method of micropropagation of hyacinths cv. Carnegie a double-phase medium has been applied. The medium was supplemented with cytokinin BA /1 mg/dm3/ and various concentrations of auxins IAA, IBA, NAA. The quality of the plantlets was evaluated on the basis of biometrical measurements as well as on the weight of fresh and the content of dry matter.

Appreciable effects of the double-phase medium containing 0.1 mg/dm3 NAA on the initiation of adventitious buds were observed. The double-phase system proved beneficial for the rates of shoots and leaves propagation, the fresh weight of explants and the length of original and newly formed leaves. In addition, the presented system is more economical than the single-phase one and gives prospects for an effective year-round production of microcuttings and/or plantlets.
