CRYOPRESERVATION OF LILY MERISTEMS
Cryopreservation may become an important tool for long term conservation of valuable lily cultivars.
We selected meristems for cryopreservation because of their genetic stability upon regrowth in normal tissue culture.
The excised meristems only survived cryopreservation after a preculture at 5°C for at least 5 days.
Preculture in the presence of dimethylsulfoxide (DMSO) followed by a direct plunge into liquid nitrogen or preculture without DMSO followed by a short treatment with cryoprotectants (DMSO and/or glycerol) and slow freezing, resulted in survival percentages of 8% or less.
