PCR ANALYSIS OF SELF-INCOMPATIBILITY ALLELES IN APPLE APPLIED TO LEAVES, SEED EMBRYOS AND IN VITRO SHOOTS

We developed a molecular technique for the diagnostic identification of 11 different self-incompatibility alleles (S alleles) in apple. The method consists of allele-specific PCR amplification of genomic DNA. Different applications of our S allele typing method are described. The S allele-specific PCR analysis was applied to leaves, seed embryos and in vitro shoots of apple. DNA templates used in the analysis are prepared either by a single-step NaOH extraction method or by a longer CTAB-miniprep method.
