STUDY ON 14C METABOLISM OF EMBRYO SEGMENTS FROM PLUM (PRUNUS DOMESTICA L.)

It is known that in plum (Prunus domestica L.) the different segments of one and the same embryo have different regenerating capacity. Those situated close to the embryo ax (first segment) give 70-100 % regeneration, while the most remote (third segment) – 0-20 %, respectively. That makes them a good model system of studying the regenerating mechanism in the traditionally difficult stone fruit species.
The aim of the present work was to investigate the differences in the 14C metabolism in regenerating and non-regenerating segments from plum embryos. Embryos from dry seeds of Stanley plum cultivar have been used in the experiments. The cotyledons were segmented into three parts, the first and the third segments were placed into nutrient medium for regenerating. The analysis was carried out following standard biochemical and radiometric procedures after fixing in hot ethanol on the 5th, 11th and 17th days.
It was established that the segments with high regenerating potential (the first segment) were characterized by rapid uptake of 14C-saccarides from the nutrient medium into the tissues and by bigger consumption for breading, compared to the third segment.
The analysis of data for the distribution of 14C in the major organic fractions and compounds showed similar tendencies in the regenerating and non-regenerating tissues. There is a clearly outlined metabolic path of sugars through amino acids and organic acids to a synthesis of high molecular compounds – nucleic acids, proteins, lipids and cellulose. Early and intensive marking with 14C of the nucleic acids and proteins was observed in both types of segments, a typical feature for active cell division. Differences between the first and the third segments were detected mainly during the second stage (12th day), when the regenerating segments showed more active metabolism of the carbohydrates uptake from the medium and more intensive synthesis of high molecular compounds compared to the non-regenerating ones.
