A COMPREHENSIVE EVALUATION OF DH-MATERIAL IN APPLE

Haploids in apple (Malus x domestica) were induced using anther and microspore culture. Flow cytometrical analysis has shown a distribution of the ploidy level from the haploid to the tetraploid state within the investigated lines of the same donor genotype. In addition to the characterization of the homozygosity state by one or two isozymemarkers per donor genotype, one apple specific microsatellite was used for molecular characterization. The analysis of anther-derived lines by the SSR marker corroborates the previous results obtained by isozymes, all alleles of the parent cultivars were also found in the progeny, but only one allele per locus was amplified in each sample. According to these analyses the androgenic regenerated shoots are homozygous. Molecular studies were carried out to investigate the evidence of markers linked to the Vf gene for scab resistance from Malus floribunda and the scab resistance gene Vr1 from ‘Russian seedling’ R12740-7A (Boudichevskaia, 2004). Out of 50 DH-lines of the cultivars ‘Remo’, ‘Rene’, ‘Releika’ and ‘Remura’ tested, 36 demonstrated bands of the corresponding markers.
