UTILIZATION OF MOLECULAR BIOLOGY METHODS IN HOP BREEDING AND MANAGEMENT IN THE CZECH REPUBLIC

The development of DNA technology has provided a number of methods, which eliminate an influence of environmental factors, to detect the differences at DNA sequence level for evaluation of hop (Humulus lupulus L.) genotypes. In our laboratory, we have used a majority of these methods (RFLP, RAPD, STS, SSR, ISSR and AFLP) for DNA fingerprinting of hop genotypes. The use of molecular methods was successfully introduced to hop breeding and management in our country. We proved that utilization of four STS primer combinations successfully and completely identified and determined Czech released varieties and new promising breeding materials. The use of STS method has been also very effective and sensitive for control of authenticity and purity of varieties in multipropagation cycle. We applied and improved AFLP, a very effective and sensitive method, for identification of somaclonal variability and genetic stability in in vitro conditions, study of inter- and intra-clonal variability, and genetic diversity of 69 actual world hop genotypes. The use of accurate and sensitive specific PCR assays has been routinely employed for detection and diagnosis of fungal, viral and viroid pathogens in naturally infected hop plants in our laboratory. The use of specific STS for marker-assisted selection (MAS) provides rapid and reliable identification of sex in F1 progenies. The use of other molecular markers for MAS will be prepared in the future. Actually, we also study the possibility of using DNA micro arrays for DNA fingerprinting and other applications. Our results are successfully being used by breeders, multipropagators and hopgrowers.
