EFFECTS OF CARBON AND NITROGEN SOURCES ON PROTOCORM-LIKE BODIES PROLIFERATION OF DENDROBIUM HUOSHANENSE BY LIQUID CULTURE
Effects of carbon sources and nitrogen sources on proliferation of protocorm-like bodies (PLBs) of Dendrobium huoshanense were investigated in liquid culture after selecting of medium types.
Among the eight basal media including full- and half-strength of MS, B5, N6 and SH, SH was the best suitable for PLBs growth.
Sucrose, glucose and fructose as carbon sources showed different influences on PLB growth.
Sucrose markedly promoted PLB growth as compared to glucose and fructose, and the optimum concentration of sucrose for PLB growth was 30 g L-1. The highest growth in medium containing glucose and fructose was only 50.9% and 51% of that in medium containing 30 g L-1 sucrose.
Higher initial concentration of sugar were favorable to dry material accumulation.
The optimum concentration of initial nitrogen was 15 mmol L-1 and NH4+ was not beneficial to growth.
