RESULTS WITH THE MICROPROPAGATION OF GALANTHUS ELWESII AND GALANTHUS NIVALIS ´FLORE PLENO´

The effect of different cytokinins, auxins and different concentrations of sucrose was examined on Galanthus elwesii bulb scale multiplication. Bulbs were desinfected with 0,3% HgCl2 propagated on a starting medium. Bulblets obtained were transplanted to BA, Kinetin, 2iP, NAA, IBA and 2,4-D containing media in different combinations. The effect of sucrose concentration was also tested. Highest number of bulblets were proliferated on 2iP + IBA containing medium, but the size of them was very small and vitrification also occurred. Bulb multiplication was in best quality and quantity on medium containing 2,0 mg/L BA + 2,0 mg/L IBA. 25% of bulblets were bigger than 5 mm in diameter, suitable for acclimatization. With the increase of sucrose concentration the number of daughter bulbs decreased, best amount was 20 g/L sucrose. Bulbs bigger than 5 mm in diameter could be acclimatized without problems, but the flowering failed to come. Dark treatment had positive effect on bulblet formation of Galanthus nivalis ‘Flore Pleno’ in the starting stage, with dark treatment 3,33 daughter bulbs were formed from one inoculum on BA and NAA containing medium.
