IN VITRO REGENERATION PLANTS OF TWO CULTIVATED TOMATO LYCOPERSICON ESCULENTUM MILL.

The regeneration technique was applied in the cultivated tomato Lycopersicon esculentum. Mill var. Justar and Nemador. The two genotypes used showed a significant differences in their aptitude for regeneration. Comparison was made on leaf and cotyledon explants cultivated on 3 different regeneration media. Shoots were obtained from seedling leaf explants of 18–21 days old, cultivated on IK basal medium supplemented with 1 mg.L-1 of Zeatin, or 1 mg.L-1 of IAA and 2 mg.L-1 of BAP. The higher regeneration rate was obtained on the media containing 1 mg.L-1 Zeatin. After elongation, shoot buds were excised and rooted with 0.1 mg.L-1 AIA. From the Justar and Nemador genotypes, leaf explants showed the most important organogenesis capacity (> 90%) than cotyledon explants. The results suggest the use of leaf explants for regeneration and transformation process though it is difficult to manipulate leaves on sterile conditions. This kind of explant is used to obtain a high level of diploid transgenic plants.
