LONG-TERM SUBCULTURE AND MAINTENANCE OF TRANSGENIC RESISTANT EMBRYOGENIC CALLUS IN LITCHI (LITCHI CHINENSIS SONN.)

The effects of subculture method, hygromycin, AgNO3, inoculation quantity and subculture cycle on the growth of litchi transgenic resistant embryogenic callus (TREC) were studied in this experiment. Meanwhile, the influences of the medium and long-term subculture on the GUS expression of litchi resistant embryogenic callus were carried out. The obtained results showed that litchi TREC could be steadily maintained for a long time when they were alternately subcultured in the MS medium containing 1.0 mg/L 2,4-D, 50 mg/L hygromycin and the MS medium supplemented with 1.0 mg/L 2,4-D, 100 mg/L inositol and 50 mg/L hygromycin.
