ANALYSIS OF LONGAN POD ISOENZYME ACTIVITY
The methods of POD isoenzyme extraction and electrophoresis were optimized.
The result showed that 3 to 6 month young leaf was suitable for POD isoenzyme.
The extraction buffer contains 0.05 mol/L phosphate buffer (pH 7.5) and 0.1% TritonX-100. POD isoenzyme was separated by polyacrylamide gel electrophoresis.
The zymogram of POD isoenzyme increased from 8 to 17 using improved POD extraction and electrophoresis methods.
The zymogram was divided into 4 regions.
Region I and III were active regions.
The zymograms in these regions were active and conserved.
Region II and IV were weak regions, and the zymograms were weak.
It was concluded that the second and third bands were distinct bands.
