DETECTION OF X-DISEASE PHYTOPLASMA IN PICEA ABIES A. DIETR. TREE IN POLAND
The detection of phytoplasma classified in phylogenetic group 16SrIII, X-disease, in a Picea abies tree with shoot fasciations and twisting in Poland is reported.
The presence of phytoplasma in needles of malformed shoots was demonstrated by nested polymerase chain reaction assay employing phytoplasma universal P1/P7 followed by fA/rA or R16F2n/R16R2 primer pairs.
Amplified cDNA of spruce isolate was sequenced (GenBank accession number EU191028). The nucleotide sequence of the spruce isolate showed more than 98,5% similarity with the corresponding fragments of sequence of phytoplasmas belonging to X-disease group, 16SrIII, now proposed to be named Candidatus phytoplasma pruni.
