IN VITRO DIRECT PLANT REGENERATION FROM LEAVES OF BLACKBERRY CULTIVAR 'BOYSENBERRY'
Plantlets were regenerated in vitro from leaves of blackberry hybrid cultivar Boysenberry. Leaflets from 2-month-old in vitro-grown plantlets on MS medium with 0.1 mg/L TDZ were used as the explants.
After 20 days of culture, adventitious buds emerged at their petioles,performance of the 2nd (from top) leaflet was best.
The 2nd leaflet from 1- to 5-month-old in vitro-grown plantlets were treated in the same way, and leaflets from 3-month-old plantlets showed best.
The 2nd leaflets of 3-month-old in vitro-grown plantlets were cultured on MS medium with 0.05 mg/L TDZ was the optimal medium for inducing adventitious buds without callus; MS medium with 1.5 mg/L KT + 0.1 mg/L NAA was the optimal medium for prolifera-tion.
The best rooting medium was 1/2 MS with 0.03 mg/L NAA.
