DIFFERENTIAL IN VITRO BEHAVIOUR OF THE SPANISH OLIVE (OLEA EUROPAEA L.) CULTIVARS 'ARBEQUINA' AND 'PICUAL'

Nodal explants from the spanish cultivars ‘Arbequina’ and ‘Picual’ showed different behaviour under in vitro conditions. In the initiation medium which included the DKW mineral and vitamins formulations 3% sucrose, 4.4 µM BAP and 0.05 µM IBA, the observed sprouting rates were 68 and 57.7% for ‘Arbequina’ and ‘Picual’ respectively. For shoot proliferation, the effects of five different media were tested. Best results for ‘Arbequina’ were obtained in a modified basal DKW medium supplemented with 2% mannitol and 5.7 µM ZR, with a 2.9 multiplication rate, while for ‘Picual’, 1.6 multiplication rate was observed in the presence of OM basal medium with 3.6% mannitol, 13.68 µM Z, 4.33 µM GA3 and 0.49 µM IBA.
