PHYLOGENETIC ANALYSIS OF SOME ANDROECIOUS GENOTYPES NATIVE TO CHINA AND RELATED DIOSPYROS SPP. USING CHLOROPLAST FRAGMENTS

Some new androecious genetic resources of Diospyros have been recently collected in Dabieshan Mountains in Central China. The taxonomic status of this germplasm is not distinguished and needs to be further exploited. Phylogenetic relationshiops of those androecious genotypes and seven species including D. kaki Thunb., D. lotus L., D. glaucifolia Metc., D. oleifera Cheng., D. rhombifolia Hemsl., D. kaki var. silvestris Mak. and Jinzaoshi (D. sp.) were revealed by four chloroplast regions including matK, trnL-trnF, trnL intron and psbA-trnH using Maximum Parsimony (MP), Maximum Likelihood (ML) and Neighbour Joining (NJ) methods. The results of sequence alignment showed that the differences just exhibited in interspecies. Meanwhile the four cpDNA fragments’ sequences of D. kaki var. silvestris Mak. were the same as D. kaki Thunb., indicating it may share a close relationship to D. kaki. In addition, five cultivars of D. kaki Thunb. involving C-PCA (Chinese origin), C-PCNA (Chinese origin), and J-PCNA (Japanese origin) owned the externally same sequence suggested that those four chloroplast regions cannot be used to distinguish the intraspecies of D. kaki Thunb. Moreover, seven androecious germplasms shared the same sequences with D. kaki Thunb. The phylogenetic tree revealed that the seven androecious germplasms were clustered with D. kaki Thunb., which implied that those androecious genetic resources should be grouped to D. kaki Thunb.
