ISOLATION OF DKADH1 GENE FROM 'NANTONG-XIAOFANGSHI' PERSIMMON AND CONSTRUCTION OF ITS PLANT EXPRESSION VECTOR
Nantong-xiaofangshi (Diospyros kaki Thunb.), a dwarf persimmon cultivar, originates in Nantong, Jiangsu Province.
An alcohol dehydrogenase 1 (ADH1) gene was isolated and named DkADH1. Bioinformatics methods were employed to analyze and predict the genetic encoding protein, secondary and tertiary structure domain, function domain of DkADH1. The results showed that the open reading frame (ORF) of DkADH1 was 1137 nucleotides encoding 379 amino acids.
It has the structure domain and function domain of the typical ADH gene.
We analyzed the evolution relationship of ADH genes among different species to construct the phylogenetic tree.
The results indicated that DkADH1 could be classified into the same group with dicotyledonous plants and was close to Solanum lycopersicum and Solanum tuberosum. The plant expression vector of pYH4215 with DkADH1 gene was constructed.
