THE EFFECTS OF MEDIUM INGREDIENTS ON SHOOT PROPAGATION AND ROOTING OF CHERRY ROOTSTOCKS IN VITRO

The effects of nutrient composition and concentrations, the concentrations of sucrose, thiamine and growth regulator concentrations were investigated on ‘Gisela 5’ and ‘Maxma 14’ rootstocks in vitro. For multiplication of cherry rootstocks, full strength MS medium containing 30 mg sucrose/L, 6 g agar/L and 25 mg thiamine/L, and supplemented with 0.5 mg BAP/L, 0.01 mg IBA/L, and 0.1mg GA3/L for ‘Gisela 5’, 0.5 mg BAP/L, 0.1 mg IBA/L, and 0.1mg GA3/L for ‘Maxma 14’ were the best medium compositions. Decreasing the nitrates concentrations induce more roots from the ‘Gisela 5’ explants. Best rooting was observed when the medium was supplemented with 0.5–1 mg IBA/L for ‘Gisela 5’ and hormone free medium for ‘Maxma 14’.
